Plasma Treated Effect of Poly (Lactic-co-Glycolic acid) (PLGA) Scaffold on Proliferation and Differentiation of Preadipocyte

Abstract
Human preadipocytes attachment to poly (lactide-co-glycolide) [PLGA] sponge was examined in vitro. This study was performed to investigate the effect of surface modification of poly (lactic-co-glycolic acid) (PLGA) scaffold by plasma glow discharge treatment on the adhesion, proliferation and adipogenesis of human preadipocytes, and not plasma glow discharge treatment. Preadipocytes were isolated from human subcutaneous adipose tissue obtained by lipectomy and liposuction. The amounts of DNA for cell proliferation rate of three-dimensional cultured group (PLGA) were higher than monolayer cultured group. At the beginning of culture, the amounts of DNA of preadipocyte in PLGA with plasma treated were higher than those without plasma treated. There was no significant difference of DNA amount between PLGA with plasma treatment and those without plasma treatment after 7 day culture. Messenger RNA of PPAR- gamma and LPL from all the preadipocyte cultured by differential medium were expressed by RT-PCR. On the 7 th and 11th day after culture, m-RNA expression of PLGA with plasma treatment were stronger than those of PLGA without plasma treatment. These results suggest that plasma treatment of PLGA sponge not only could increase the adhesion property and proliferation of preadipocytes, but also provide suitable environment for differentiation of preadipocytes.
